Your body makes its own cannabis-like compounds. Two of them are particularly important: 2-AG and anandamide. Understanding the difference between them helps you understand how the endocannabinoid system actually works.
2-AG: The Workhorse
2-arachidonoylglycerol (2-AG) is the most abundant endocannabinoid in your body. It's the workhorse of the system, constantly being produced and broken down to regulate moment-to-moment neural activity.
2-AG is primarily involved in retrograde transmission—the feedback mechanism that prevents neurons from over-firing. When a neuron is getting too much stimulation, it releases 2-AG to tell the sending neuron to back off.
Think of 2-AG as your brain's real-time volume control.
Anandamide: The Bliss Molecule
Anandamide gets its name from the Sanskrit word "ananda," meaning bliss. It was the first endocannabinoid discovered, and it's associated with mood, motivation, and the runner's high.
Unlike 2-AG, anandamide is produced on demand in response to specific stimuli. It's more like a special messenger than a constant regulator. Exercise, chocolate, and social bonding all boost anandamide levels.
Dr. Bob Melamede often pointed out that anandamide is what makes us feel good when we do things that are good for us. It's evolution's reward system, encouraging behaviors that promote survival and adaptation.
Different Roles, Same System
Both endocannabinoids work through the same receptors (CB1 and CB2), but they have different affinities and different roles. 2-AG is a full agonist at CB1 receptors; anandamide is a partial agonist. This means they produce different effects even though they're working through the same system.
